    摘要:

    通過添加Vc和外加磁場處理,,研究蓮藕多酚氧化酶(PPO)反應(yīng)動力學的變化規(guī)律,。以鄰苯二酚為底物,,蓮藕PPO褐變反應(yīng)動力學符合米氏方程所描述的單底物酶促反應(yīng)動力學,,Km為0.775mol/L,,Vmax為11.150U/min,。Vc對PPO有較強的抑制作用,,反應(yīng)動力學參數(shù)Km為0.081mol/L,,Vmax為3.243U/min,,呈現(xiàn)反競爭性抑制,,表現(xiàn)為褐變出現(xiàn)滯后,隨Vc濃度的增大滯后時間逐漸增長,,且呈現(xiàn)S型趨勢,,并建立了相應(yīng)的反應(yīng)方程。不同磁場強度下,,隨著處理時間的變化,,PPO活性呈現(xiàn)波動性。3.17A/m磁場強度下處理4h的酶液其反應(yīng)動力學曲線表現(xiàn)為S型,,并建立了相應(yīng)的反應(yīng)方程,。

    Abstract:

    The change law of reaction dynamics was experimental studied through adding Vc to lotus root polyphenol-oxidase and dealing with lotus root polyphenol-oxidase (PPO) by magnetic field. The experimental results indicated that the reaction kinetic of enzymatic browning was accordant with Michaelis equation with catechol substrate, which Km and Vmax were 0.775 mol/L and 11.150U/min respectively. Vc had an inhibiting effect on lotus root PPO, which Km and Vmax were 0.081mol/L and 3.243U/min respectively. The browning was lagged and the lag time increased gradually with the rise in the concentration of the inhibitors, which was an anticompetitive inhibition. The PPO activity expressed a fluctuation at different magnetic field strengths and times. At 3.17A/m magnetic field strength for 4 hours, the reaction kinetics curve was a wave-type curve. The kinetic equations were established respectively.
