    摘要:

    借助Design-expert統(tǒng)計(jì)系統(tǒng),采用中心組合試驗(yàn)法對(duì)真姬菇菌絲體多糖的堿提取工藝進(jìn)行了優(yōu)化研究。結(jié)果表明,以提取溫度、提取時(shí)間,、液料比、乙醇用量四因素為自變量,以多糖提取率為因變量,,回歸得到二次多項(xiàng)式模型極顯著,模型校正決定系數(shù)為0.9495,,相關(guān)系數(shù)為0.9847,。當(dāng)提取溫度為73~76℃、提取時(shí)間為2.6~3.5h,、液料比為18.25~25mL/g,、乙醇用量為提取液體積的2.5~3.5倍時(shí),多糖提取率最低為4.45%,。當(dāng)提取溫度為76℃,、提取時(shí)間3.24h、液料比為23mL/g,、乙醇用量為提取液體積的3.5倍時(shí),,多糖提取率達(dá)最高值

    Abstract:

    5.23%。Based on design-expert statistic system, extraction conditions including temperature, time, the ratio of alkaline solution and mycelium, and the volume ratio of alcohol and alkaline solution were optimized for mycelium polysaccharide by center composition design. The results showed that quadratic model was the best model to describe the relationship between the extraction rate of polysaccharide and the factors. R-squared adjusted value of the model was 0.9495, and the simulated coefficient R was 0.9847.By response surface methodology (RSM), we found that when temperature, time, the ratio of alkaline solution and mycelium, and the volume ratio of alcohol and alkaline solution respectively were 73~76℃, 2.6~3.5h, 18.25~25mL/g and 2.5~3.5, the lowest extraction rate of polysaccharide was 4.45%. When temperature, time, the ratio of alkaline solution and mycelium, and the volume ratio of alcohol and alkaline solution separately were 76℃, 3.24h, 23mL/g and 3.5, the extraction rate of polysaccharide would reach 5.23%, a highest value in our investigation extension. 
