    雌二醇是一種典型的雌激素,易通過食物鏈影響人體健康。因此,探索簡單、快速、靈敏的雌二醇檢測技術(shù)及方法很有必要。以納米金比色分析為基礎(chǔ),,以雌二醇特異性適配體(長度為76mer)為傳感探針,在優(yōu)化試驗(yàn)條件的基礎(chǔ)上,,提出一種基于核酸適配體的雌二醇納米金比色傳感檢測方法,,并分析了對牛奶和雞蛋中雌二醇檢測的可行性。結(jié)果表明,,當(dāng)納米金粒子與核酸適配體濃度比為1∶11000,、在室溫孵育30min后,在NaCl濃度為24mmol/L的條件下,,水體系的A650/A530與雌二醇濃度間呈良好的線性關(guān)系(R2=0.915),,線性區(qū)間為0.25~0.60nmol/L,檢測限為0.233nmol/L,,具有良好的特異性,。該方法可實(shí)現(xiàn)對牛奶和雞蛋樣品中雌二醇的檢測,對牛奶和雞蛋樣品的線性檢測區(qū)間分別為0.5~0.8nmol/L和0.3~0.9nmol/L,,檢測限分別為0.183nmol/L和0.185nmol/L,,加標(biāo)回收率分別為101.6%~105.4%和97.97%~115.30%,相對標(biāo)準(zhǔn)偏差小于6%,。該方法具有較高的檢測靈敏度和特異性,,且簡單可行,為牛奶,、雞蛋樣品中雌二醇的快速檢測提供了新思路,。

    Abstract:

    As a typical estrogen, 17β-estradiol can easily affect human health through accumulation in animal food. Therefore, the establishment of a simple and sensitive technique for rapid analysis of 17β-estradiol is of great practical significance. On the basis of the gold nanoparticles colorimetric analysis, a gold nanoparticles colorimetric detection method for 17β-estradiol based on aptamer was designed, using the 17β-estradiol aptamer as the sensing probe. The test conditions were also optimized, and the feasibility of this method for the detection of 17β-estradiol in milk and eggs was analyzed. Under the optimized conditions, the A650/A530 of water system showed a good linear correlation with the concentration of 17β-estradiol (R2=0.915) with a linear range from 0.25nmol/L to 0.60nmol/L, and the detection limit was 0.233nmol/L. The proposed method also illustrated an excellent specificity to 17β-estradiol, when the ratio of gold nanoparticles to aptamer was 1∶11000 which were incubated at room temperature for 30min, and under the condition of 24mmol/L NaCl. The method can also be applied successfully for the detection of 17β-estradiol in milk and egg samples. The linear range were 0.5~0.8nmol/L (milk) and 0.3~0.9nmol/L(eggs) with the detection limit of 0.183nmol/L and 0.185nmol/L, respectively. The recovery were 101.6%~105.4% (milk) and 97.97%~115.30% (eggs), respectively. Meanwhile, the relative standard deviation was less than 6%. With the advantage of higher sensitivity and specificity, the simple and practical method can provide a way for the rapid detection of 17β-estradiol in milk and egg samples.
